miR-21 如何促进毛囊神经嵴干细胞向许旺细胞分化? 毛囊神经嵴干细胞可分化为许旺细胞,但分化过程中相关的调节机制目前尚未完全清楚。中国吉 林大学口腔医院倪宇昕博士所在团队,从人毛囊中分离的神经嵴干细胞转染 miR-21,定向诱导为许 旺细胞,其中的 miR-21 在神经嵴干细胞分化为许旺细胞的过程中表达逐渐升高。转染 miR-21 激动 剂 agomir-21 后,干细胞的分化能力增强。进一步研究发现,在神经嵴干细胞中敲除 SOX2 基因可显 著逆转 antagomir-21 抑制干细胞分化为许旺细胞的能力,miR-21 可在不影响 SOX2 mRNA 水平的 情况下通过与其 3’-UTR 段结合而下调 SOX 蛋白表达。结果表明 SOX2 是 miR-21 的作用靶点, miR-21 可通过与 SOX2 mRNA 3’ -UTR 段结合下调其蛋白表达, 促进神经嵴干细胞向许旺细胞分化, 发挥修复周围神经的作用。相关文献发表于 《中国神经再生研究 (英文版) 》 杂志 2014 年 4 月第 8 期。

How does miR-21 promote the differentiation of hair follicle-derived NCSCs into SCs? Hair follicle-derived neural crest stem cells (NCSCs) can be induced to differentiate into Schwann cells (SCs). However, the underlying regulatory mechanism during cell differentiation remains poorly understood. Dr. Yuxin Ni and her team, Hospital of Stomatology, Jilin University, China isolated NCSCs from human hair follicle and induced them to differentiate into SCs. MicroRNA (miR-21) expression was gradually increased during the differentiation of NCSCs into SCs. After transfection with the miR-21 agonist (agomir-21), the differentiation capacity of NCSCs was increased. Without compromising SOX2 mRNA expression, miR-21 down-regulated SOX protein expression by binding to the 3’-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the NCSCs significantly reversed the miR-21 antagonist inhibition of NCSCs differentiating into SCs. The results suggest that SOX-2 was an effective target of miR-21 and miR-21 promoted the differentiation of NCSCs into SCs through down-regulating SOX protein expression by binding to the 3’-UTR of SOX2 mRNA. These results were published in Neural Regeneration Research (Vol. 9, No. 8, 2014).
